Carmel Napier, who has led the force since 2011, will be temporarily replaced next week while a replacement is sought.
After completing a degree in English and Music from Nottingham University, Ms Napier joined Hertfordshire Police in 1983, staying for 17 years before moving to become deputy superintendent at North Yorkshire Police. She left to join Essex Police as an assistant chief constable in 2006 and moved to Gwent as deputy chief constable two years later.
Gwent PCC Ian Johnston told the BBC Ms Napier had wished to retire following her completion of 30 years in the police, adding he had accepted her wishes.
Gwent Police, Mr Johnston and Ms Napier are unavailable for further comment.
